Darrell Cobbins, 30, was arrested on Tuesday in the 16500 block of South Kedzie in Markham, Illinois.
William Cooksey, 37, was arrested on Thursday, in the 8700 block of South Sangamon Avenue.
Chicago police said the victim was a passenger in the vehicle when someone in a dark-colored SUV fired shots.
He was taken to Christ Hospital, where he was pronounced dead.
