"He let slip that he knew the place where this person was buried," Gen. Cesar Guardia told The Associated Press.
Guardia said that he didn't know how seriously to take Van der Sloot's statement given his history of dubious claims about the 2005 disappearance in Aruba of the Alabama teen.
Van der Sloot remains the lone suspect in the case. His latest confession arose during questioning for the murder of another young woman, Stephany Flores.
Peruvian police say he has admitted killing Flores because she stumbled onto secrets relating to Holloway that were stored in his laptop.
The bloody and battered body of the 21-year-old woman from Lima was found in a hotel room listed under Van der Sloot's name. Police believe she was murdered May 30, five years to the day that Holloway disappeared during an Aruba holiday.
On Thursday, Van der Sloot, sheathed in a bulletproof vest, was moved across downtown Lima to a cell at the prosecutor's office as officials prepared to file charges in the killing of Flores, who police say he met playing poker at a casino three days earlier.
