State Police say a car was heading south in the northbound lanes and collided with another vehicle around 4:15 a.m. just before the Route 3 on ramp.
Troopers immediately shut down the northbound side of the highway near the Burlington line.
The drivers of both cars were rushed to the Lahey Clinic in Burlington in critical condition.
"The two vehicles involved are a 2008 Hyundai Accent and a 2007 Chevrolet Cobalt; at this time, the investigation into which vehicle was going the wrong way is ongoing," State Police spokesman Dave Procopio said in a statement.
No names have been released, but Procopio said the driver of the Hyundai was a 20-year-old woman from Wayland. The Cobalt driver was a 27-year-old man from Dracut.
A breakdown lane was opened for traffic to slowly get by, but there were major delays in the area for the morning rush hour.
All lanes were re-opened by 7:15 a.m.
No other information is available at this point in the investigation.
